I'm not so sure your problem has anything to do with the transactional cache. I have imported a little over 1000 users into the Alfresco repository and noticed a major slow down in user lookup. Stepping through the code I've found that the Alfresco JCR implimentation has some major scaling issues. I started a thread on the issue here but haven't gotten much response.
